No one has ever said for sure if the Lich's key you get from Adrianna opens that lock. However, if you have a Traps & Locks skill of at least 7 and a level 5 lockpick, you can make it in. You can purchase a high-level lockpick from the Thieves' guild.
It's just random treasure behind that door, so it's really not worth it. Sometimes you can get good stuff, though.
